一、背景描述：Lycopersicon esculentum (Tomato) Lectin (LEL, TL), DyLight 649, is a bright deep-red/far-red LEL-fluorophore conjugate.

It is a valuable tool in studies of rodent tumor angiogenesis and the tracing of neovascular development in xenograft models and brain research.
LEL-DyLight 649 can be applied directly to tissue sections, or it can be introduced through intravascular perfusion for fluorescence detection.

LEL is a very stable single-subunit glycoprotein containing ∼50 percent arabinose and galactose and may form multimeric aggregates in solution.
It is provided at a 1 mg/mL concentration in 10 mM HEPES, 0.15 M NaCl, pH 7.5, 0.08% sodium azide, 0.1 mM Ca2+. If a precipitate forms upon long-term storage, warm to 37°C.
LEL 是一种非常稳定的单亚基糖蛋白国融资管，含有约 50% 的阿拉伯糖和半乳糖，可在溶液中形成多聚体聚集体。
它以 1 mg/mL 浓度在 10 mM HEPES、0.15 M NaCl、pH 7.5、0.08% 叠氮化钠、0.1 mM Ca2+ 中提供。如果长期储存时形成沉淀物，请加热至 37°C。

二、荧光检测与染色问题（一）荧光参数
激发波长：655 nm，发射波长：670 nm（远红光），可用 Cy5/深红滤光片 检测。
染料结合量优化，几乎不含游离荧光素，但仍需避光操作以防淬灭。
（二）常见问题与解决
低染色效率：
可能因反应温度过低（需≥37°C）或时间不足，优化孵育条件。
高背景信号：
未充分冲洗血管（增加PBS洗涤次数）；
游离染料残留（用层析柱纯化或增加含 0.5% Tween-20的PBS 洗涤）；
非特异性结合（用 5% BSA 封闭组织切片）。
